The SIU is led by the Director and comprised of approximately 41 civilian investigators (ie., non-police officers). There are 3 Investigative Supervisors and 12 full-time Investigators who work out of the SIU's office in Mississauga, with the remainder stationed across the province and deployed on an as-needed basis. This arrangement ensures prompt response times regardless of where in the province an incident occurs. The investigative staff includes individuals who are experts in traffic accident investigations and collision reconstruction.

The SIU also has its own in-house Forensic Identification Section, with capacity to assist in all manner of forensic identification work. The team is led by 2 full-time Forensic Identification Supervisors and 10 as-needed Forensic Identification Technicians.
The work of the office is further assisted by an Executive Officer, Legal Counsel, Administrative Manager, Communications Manager, Training Coordinator, and an administrative staff composed of transcribers, a central registry clerk, a budget and inventory clerk, an information technology systems analyst and administrative assistants.
